What type of email account do you use in Outlook (POP, IMAP, MAPI or Exchange)?
Step: 1
You may start Outlook in application safe and check if you are able to create signature in Outlook
Click ‘Start’, Type ‘outlook /safe’ (without quotes) in search programs and files box above Start and press enter.
If Outlook works fine as expected, you may disable Outlook add-in using the link below and check if it helps:

You may start the computer in clean boot mode this would disable all the non-Microsoft applications and the antivirus which may be one of the reason for signature issue.
